Endothelial ACKR3 drives atherosclerosis by promoting immune cell adhesion to vascular endothelium

چکیده

Background and Aims : Genome wide association studies revealed a strong between the CXCL12 gene locus cardiovascular diseases, highlighting its receptors CXCR4 atypical chemokine receptor-3 (ACKR3) as targets for CVD research. Although vascular is well studied in atherosclerosis, exact role of ACKR3 remains elusive. Therefore, aim this project to decipher atherosclerosis.

Heparanase induces inflammatory cell recruitment in vivo by promoting adhesion to vascular endothelium.

Heparanase (HPSE1) is known to be involved in mechanisms of metastatic tumor cell migration. This enzyme selectively cleaves heparan sulfate proteoglycans (HSPG), which are ubiquitously expressed in mammals and are known to be involved in regulating the activity of an array of inflammatory mediators. In the present study, we have investigated the effects of human recombinant heparanase, the ina...

Androgen exposure increases human monocyte adhesion to vascular endothelium and endothelial cell expression of vascular cell adhesion molecule-1.

BACKGROUND Male sex is an independent risk factor for coronary artery disease. Owing to the importance of monocyte adhesion to endothelial cells in the development of atherosclerosis, we hypothesized that androgens might promote this process. We therefore studied the effects of the nonaromatizable androgen dihydrotestosterone (DHT) on human monocyte adhesion to human endothelial cells and on en...

Vascular endothelial growth factor enhances cancer cell adhesion to microvascular endothelium in vivo.

To investigate whether vascular endothelial growth factor (VEGF) enhances cancer cell adhesion to normal microvessels, we used in vivo video microscopy to measure adhesion rates of MDA-MB-435s human breast cancer cells and ErbB2-transformed mouse mammary carcinomas in the postcapillary venules of rat mesentery. An individual postcapillary venule in the mesentery was injected via a glass micropi...

Endothelial-to-mesenchymal transition drives atherosclerosis progression.

The molecular mechanisms responsible for the development and progression of atherosclerotic lesions have not been fully established. Here, we investigated the role played by endothelial-to-mesenchymal transition (EndMT) and its key regulator FGF receptor 1 (FGFR1) in atherosclerosis. In cultured human endothelial cells, both inflammatory cytokines and oscillatory shear stress reduced endothelia...

Adhesion to the Vascular Endothelium Cell Adhesion Molecules Mediate Radiation-induced Leukocyte

The predominant early histological changes in irradiated tissues are edema and leukocyte infiltration. Cell adhesion molecules (CAMs) are required for the extravasation ofleukocytes from the circulation. To study the role of CAMSin the pathogenesis of radiation-mediated inflammation, we quantified the expression of P-selectin, E-selectin, intercellular adhe sion molecule-i (ICAM-1), and vascula...
